摘要:

Most of the advancements made in the past two decades in cancer radiation therapy are largely based on the fundamental hypothesis that radiation causes cell death and thus tumor control or normal tissue toxicity through DNA double strand breaks. However, new research have challenged the hypothesis and elucidated that in addition to causing cellular DNA damage, localized radiation also initiates cascades of complex downstream biological responses in tissue that extend far beyond where radiation dose is deposited and beyond cell killing. The new multidisciplinary research show that massive radiation dose may not lead to normal organ function damage as long as the radiation is finely fractionated in space; locally irradiated dying tumor cells may release tumor antigens to enhance systemic anti-cancer immune response in cancer patient; targeted irradiation to solid tumor may increase the migration of tumor cells in bloodstream, the seeds of potential metastasis; and microbeam radiation significantly modulated tumor microvasculature and that can be used to drastically enhance tumor delivery of nanoparticle-encapsulation cancer drugs.
